Høgh, Celtic's record signing this summer, has been out since the second leg of their Champions League play-off defeat by LASK on 25 August.
Tierney missed the recent wins over Aberdeen and St Mirren but returned to training on Tuesday.
"We've had Kasper back the last couple of days and Kieran did a little bit of work with us this morning, so we'll have a look to see how they are," O'Neill said.
Asked if the pair could play some part, he added: "Yeah, obviously Kasper's had the extra day, we'll see how they are."
Høgh has scored three goals in five appearances since joining from Bodo/Glimt, while fellow summer signing Camilo Duran has impressed in his absence, scoring six times in eight games.
Celtic will be without winger Yang Hyun-jun, who is representing South Korea at the Asian Games. O'Neill called him a "big miss".
Celtic travel to Rangers on Sunday, but O'Neill insists Wednesday's game is the priority.
"We have to try and defend a title. We have to try and win a football match," he said.
